CRISPR Library Screening: What is it?
CRISPR library screening technology is a biological tool based on the CRISPR/Cas9 system, designed for high-throughput gene function research. The origin of this technology can be traced back to the invention of CRISPR/Cas9, a revolutionary gene editing tool initially used for single gene editing. As research progressed, scientists discovered that by integrating appropriate sgRNA sequences, multiple genes could be intervened or edited simultaneously, leading to the birth of CRISPR library screening technology.
Through high-throughput CRISPR sgRNA library screening, high-throughput gene knockout is achieved, obtaining cells with different gene edits, including gain of function (GOF) and loss of function (LOF), and exploring the biological significance of these genes. CRISPR library screening technology has been widely applied in fields such as gene function, disease mechanisms, drug target identification, and tumor immunology. It not only deepens our understanding of biological processes but also promotes continuous breakthroughs and innovations in disease treatment and biotechnology.
Currently, there are two forms of CRISPR library screening: pooled library and arrayed library. EDITGENE provides services for both types of libraries. Although arrayed libraries are becoming increasingly popular, pooled libraries are cost-effective, easy to operate, and suitable for in vivo studies, allowing for more comprehensive genome coverage. Infected cells can be tested after long-term culture. Therefore, pooled libraries are commonly used for high-throughput screening amongst CRISPR libraries.
